Thin-layer Chromatography of Acid-labile Cobalamins

Acid-labile, axial ligand-substituted cobalamins were separated by thin-layer chromatography using (TLC) on cellulose under slightly basic conditions. The compounds were dissolved in 0.5% concentrated NH4OH and applied to 20 cm x 0.1 mm microcrystalline cellulose plates. Ascending thin-layer chromatography was performed at ambient temperature in the dark to a distance of approx.10 cm, and the solvent was BuOH-EtOH-H2O (10:3:7) containg 0.5% concentrated NH4OH. The compounds were visualized by their intense red color.
