Preparation of Ruthenium(II) and RutheČnium(III) Myoglobin and the Reaction of Dioxygen, and Carbon Monoxide, with Ruthenium(II) Myoglobin

Ru myoglobins were prepared by the reconstitution of horse heart apomyoglobin with either Ru(II) or Ru(III) mesoporphyrin IX (MpIX) derivatives. The Ru(II) and -(III) myoglobins (RuMb and RuMb+) contained 1 Ru porphyrin/heme binding site; the species were readily interconverted using dithionite for reduction and Br2 for oxidation. RuMb bound CO to give the known carbonyl complex. Reversible oxygenation occurred readily with protein-free Ru(II)(MpIX) species in DMF but RuMb in phosphate buffer was irreversibly oxidized by O2 to give RuMb+ via an outer sphere electron transfer mechanism.
